B.SIGHT™ – Sequencing Single-cell Genomes of Uncultivated Bacteria in the Human Oral Microbiome

Uncultivated bacteria in the human oral microbiome remain largely unexplored, offering opportunities for new discoveries. In this study, the B.SIGHT enabled label-free single-cell isolation with image-based detection, followed by miniaturized MDA that reduced reaction volumes and costs. Subsequent 16S rRNA sequencing allowed taxonomic classification, and because all steps were performed in standard microwell plates, the workflow scaled efficiently to thousands of organisms without cultivation.

The B.SIGHT was essential for precise and cost-effective sequencing of single-cell genomes in the oral microbiome.
